چکیده

The incidence of cleft lip in CL/Fr mouse is 18-2500. The purpose of this study is to clarify the cause of cleft lip in CL/Fr mouse using whole embryo culture system which is a useful technique in studying the development of mammalian embryos in vitro. We examined the incidence of cleft lip in CL/Fr and F, mouse embryo (male CL/Fr x female C57BL/6) in vitro. The embryos were explanted on day 10 of gestation (plug day=0) and were cultured during the period of primary palate formation for 40-48 hours. The non-CL (P) producing strain C57BL/6 embryos were used as control. The cleft lip was observed in 84. 4% of the cultured CL/Fr embryos, while C57BL/6 and F1 embryos did not show any cleft lip in utero and in vitro. The incidence of cleft lip in CL/Fr mice showed variations among litters in vtero. However, in the cultured mouse embryos, there was a little variation in the incidence of cleft lip. These results might suggest that all embryos of CL/Fr mice have a genetic predisposition to cleft lip and that the gene (s) related to cleft lip in CL/Fr mouse is (are) recessive against the allele of C57BL/6 strain.

Molecular contribution to cleft palate production in cleft lip mice

Cleft palate following cleft lip may include a developmental disorder during palatogenesis. CL/Fr mice fetuses, which develop cleft lip and palate spontaneously, have less capability for in vivo cell proliferation in palatal mesenchyme compared with CL/Fr normal fetuses. In order to know the changes of signaling molecules contributing to cleft palate morphogenesis following cleft lip, the mRNA ...

Hyperoxia and hypoxia in pregnancy: simple experimental manipulation alters the incidence of cleft lip and palate in CL/Fr mice.

This paper describes alterations in the incidence of cleft lip and palate in CL/Fr mice subsequent to experimental manipulation of maternal respiratory oxygen levels during a critical period of pregnancy. Only a few previous studies have shown that the incidence of some "genetically determined" malformations in mammals can be decreased by environmental procedures. In addition to demonstrating a...

Suppression of developmental anomalies by maternal macrophages in mice

We tested whether nonspecific tumoricidal immune cells can suppress congenital malformations by killing precursor cells destined to cause such defects. Pretreatment of pregnant ICR mice with synthetic (Pyran copolymer) and biological (Bacillus Calmette-Guérin) agents significantly suppressed radiation- and chemical-induced congenital malformations (cleft palate, digit anomalies, tail anomalies,...

Cleft lip and Palate: A 30-year Epidemiologic Study in North-East of Iran

Introduction: Cleft lip and palate are among the most common congenital anomalies worldwide. This study was conducted in order to explore the incidence and related factors of cleft lip and/or palate (CL/P) among live births in Mashhad, North-Eastern Iran.   Materials and Methods: In this cross-sectional study, records of 28,519 infants born between March 1982 and March 2011 at three major hospi...

Distinct functions for Bmp signaling in lip and palate fusion in mice.

Previous work suggested that cleft lip with or without cleft palate (CL/P) is genetically distinct from isolated cleft secondary palate (CP). Mutations in the Bmp target gene Msx1 in families with both forms of orofacial clefting has implicated Bmp signaling in both pathways. To dissect the function of Bmp signaling in orofacial clefting, we conditionally inactivated the type 1 Bmp receptor Bmp...
